For decades, researchers have devoted great efforts to studying the tumor suppressor gene Trp53, however, our understanding of Trp53 regulation has never reached either organism-scale or single-cell resolution. Here, we performed spatial RNA analysis to confirm the Trp53 KO-induced tumor initiation states. Tissues of Trp53-/- and Trp53+/+ mice at different ages were isolated and embedded in Optimal Cutting Temperature compound (OCT, Sakura Tissue-TEK) on dry ice, followed by cryosectioning, fixation, tissue transferring to slides, spatial cDNA library construction, high-throughput sequencing, and bioinformatics analysis.
