Single-cell transcriptome analysis of lineage-marked prostate primary tumors and metastases from the NPK mouse model (Nkx3.1CreERT2/+; Ptenflox/flox; KrasLSL-G12D/+; R26R-CAG-LSL-EYFP/+) Overall design: Male mice of a a predominantly C57BL/6 background were induced to form tumors at 2-3 months of age by administration of tamoxifen using 100 mg/kg (in corn oil) once daily for 4 consecutive days. Following tamoxifen-induction, mice were monitored daily and euthanized when their body condition score was <1.5, or when they experienced body weight loss >20% or signs of distress, such as difficulty breathing or bladder obstruction. Single-cell transcriptomic analysis of bulk tissues was done on two matched primary tumors and macrodissected bone metastases from 2 independent mice.
