The MYCN oncogene is amplified in human neuroblastomas and it has been involved in the repression of transcriptional responses to interferon gamma. This may have important implications for the immunee phenotype of neuroblastomas and the design of immunotherapeutic inhibitors. Targeted modulation of immune and interferon signaling represents a potential strategy to overcome these barriers. We treated a panel of eight human neuroblastoma cell lines - four MYCN-amplified and four MYCN-non-amplified cell lines - with interferon gamma alone or in combination with UNC-0638, a small molecular inhibitor of the EHMT2 and EHMT1 (also known as G9a and GLP) H3K9 histone methyltransferases. Total RNA was isolated and analyzed by 3'mRNA-Seq.
