Ovarian Cancer Ascites Inhibits Transcriptional Activation of Natural Killer Cells partly through MUC16

Purpose:To investigate the effect of ascites of ovary cancer on human NK cells at the genome-wide level. Overall design: Patient ascites NK cells were purified directly from ascites of human ovary cancer patients. Human peripheral blood NK cells were isolated from freshly prepared PBMCs taken from healthy donors. Cells were treated with either medium or human ascites of overy cancer for 24 hrs. Cells were lysed in RLT lysis buffer and total RNA extracted using the RNeasy micro kit for RNAseq.
