资源简介:
The locus coeruleus noradrenergic (LC-NA) system plays an important role in organizing a physiological response to acute stress. However, if and how the LC affects stress-mediated transcriptomic changes in the brain is still barely understood. Here, we extensively characterize LC mediated transcriptomic response during acute stress in the hippocampus. Combining for the first time bulk mRNA-sequencing and selective chemogenetic and optogenetic manipulations of the LC-NA system This repository contains data from 4 different experiments: First experiment: chemogenetic activation of the locus coeruleus (LC) followed by sequencing of the ventral hippocampus (vHC) and the dorsal hippocampus (dHC) 45min after activation. Two-group design (hM3Dq virus vs control virus). Second experiment: optogenetic activation of the LC followed by sequencing of the vHC 45 and 90min after stimulation. Three groups, control virus, Chr2 virus after 45min and ChR2 virus after 1h30min. Third experiment: Optogenetic stimulation of the LC at different stimulation frequencies (Sham stimulation, 3Hz, 5Hz, 15Hz) followed by sequencing of the vHC 45min after stimulation. Fourth experiment: Selective Retrograde optogenetic stimulation of hippocampus-projecting LC neurons only followed by sequencing of the vHC 45min after stimulation. Three groups, homecage control without virus injection/stimulation, control virus and ChR2 virus.
