SIX1 is a target of MLL-AF9 and MOZ-TIF2 fusions in murine models of AML. SIX1 is also expressed in a subset of human AML cell lines. Being a transcription factor we have studied the functional loss of SIX1 in this cell line. Overall design: ChIP sequencing of 1x10^8 formaldehyde crosslinked WT THP1 cells, using IgG as an isotype control. CRISPR CAS9 generated THP1 cells were also transduced with two guides targeting exon 1 and 2 of the SIX1 gene. Cells were sorted for guide expression and sequenced at 24, 48 and 72hrs to understand the dynamic gene changes upon SIX1 loss.
