Sertoli cells play a crucial role in supporting spermatogenesis in the testes. In this study, we aimed to investigate the molecular changes in Sertoli cells after the knockdown of Dhx37, a gene involved in RNA splicing and gene regulation. siDhx37 was transfected into the mouse Sertoli cell line, followed by RNA-seq analysis at Day 0 and Day 3 post-transfection. The results reveal that Dhx37 depletion affects the expression of key genes involved in Sertoli cell function, spermatogenesis, and cell cycle regulation. Differentially expressed genes (DEGs) were identified, highlighting pathways related to cellular processes, including cell proliferation, apoptosis. Our findings suggest that Dhx37 may play an important regulatory role in Sertoli cells by modulating the expression of genes necessary for their function in male reproduction. Overall design: RNA-seq profiling of wildtype mouse Sertoli cells and their knockdown derivatives (siDhx37) at Day0 and Day3 post-transfection.
