A novel model of pulmonary KPC infection reveals critical roles of NK cell and group 3 ILC cytokines in host resistance

Both NK cells and group 3 ILCs play key roles and that effector cytokines made by these cells can provide immunoprophylaxis against Carbapenem resistant K. pneumoniae infection. Overall design: Our data had indicated that 12 hours post pulmonary infection of ST258 at 10e6 CFU reveals similar bacterial clearance between C57BL/6 and Rag2-/- mice, and therefore we harvested lung tissues to obtain single cell suspension for single cell RNAseq at this time point because any differences in gene expression would not be due to differences in lung bacterial burdens. Similarly, about the experiment for comparison between PBS and FK506 treated C57BL/6 mice, we decided to analyze at 24 hours after infection based on the same bacterial burden between the groups.
