Tumor infiltrating T cells are a diverse and heterogeneous population. We used single cell RNA sequencing (scRNA-seq) to analyze gene expression changes in tumor-specific T cells that were sufficient for or lacked PSGL-1 (gene: Selplg). In vitro activated congenically marked PSGL-1+/+ or PSGL-1-/- OT-I CD8+ T cells were adoptively transferred into separate C57BL/6 host mice bearing B16-OVA tumors. 6 days after transfer, donor OT-I cells were isolated by fluorescence-activated cell sorting (FACS) based on donor marker (CD90.1 or CD45.1) expression and analyzed using scRNA-seq (10X genomics). Biological replicates were pooled prior to sorting to generate one sample per condition.
