Immunzition of Salp14-C mRNA and lipid nanoparticles elicited multiple immune response to tick challenge in guinea pig skin

In this study, we immunized guinea pigs with mRNA of Salp14 C terminus and lipid nanoparticles (LNP), the animals produced high titers of IgG, and developed moderate erythema during tick challenge, the transcriptomes of skin at tick bite sites enriched multiple immune response pathways, which might be involved in erythema development. Overall design: we immunized two guinea pigs with Salp14-C mRNA-LNP and two guinea pigs with luciferase mRNA-LNP (control) for three times with 3-week interval, then performed tick challenge with 30 nymph/animal, then did skin biopsies at tick biste sites after all ticks detached. Total RNAs were extracted, and cDNA libraries were consructed, and bulk RNA sequencing performed.
