The authors used a co-culture of primary human brain microvascular endothelial cells (HBMEC) with Plasmodium falciparum material, recombinant PfEMP1 or lysates from infected erythrocytes (IE), and measured barrier function by Trans Endothelial Electrical Resistance (TEER). A selection of Protease-Activated Receptor-1 (PAR1) inhibitors was tested for their ability to reverse the P. falciparum and thrombin induced decrease in barrier function. The TEER data is from an initial screen with 10 PAR1 inhibitors and the human umbilical vein endothelial cell line EA.hy926 followed by more detailed testing of 4 selected inhibitors and HBMEC. In addition, the effect of IE or IE lysate on EPCR expression on HBMEC was determined by flow cytometry.
