Given the high druggability of kinases, we utilized a targeted library focused specifically on the kinome for in vivo CRISPR/Cas9 knockout (KO) screening Overall design: To conduct the screen, lentiviruses carrying the kinome-targeting sgRNAs were used to infect mouse CT26 cancer cells. Following a 7-day selection process to remove uninfected cells, the transduced cells were divided into three groups: in vitro two-dimensional (2D) culture, and in vivo experiments through subcutaneous injection to establish xenograft tumors in either immunodeficient BALB/c nude mice (BALB/c Nude) or immunocompetent wild-type BALB/c mice (BALB/c WT).
