In this study, we aimed to investigate the differentiation dynamics of megakaryocyte development, utilizing human umbilical cord blood (UCB) cells We cultured human umbilical cord blood (UCB) cells to generate megakaryocytes by first isolating mononuclear cells from UCB using density gradient centrifugation, then plating the cells in a serum-free medium supplemented with thrombopoietin (TPO) to promote megakaryocyte differentiation, maintaining the cultures at 37°C in a humidified atmosphere with5% CO2, and periodically assessing cell morphology and surface markers using flow cytometry to confirm the generation of megakaryocytes
