DNA mismatch repair deficiency and longstanding inflammatory bowel disease predispose to colorectal cancer. Using a novel double knockout mouse (MSH2 loxP/loxPVil-cre x IL-10 -/-) we could demonstrate that proximal colonic tumourigenesis in mismatch repair deficient mice is aggravated under chronic inflammation. Tumour development in the proximal colon was accompanied by bacterial invasion into tumour crypts. These MSH2 loxP/loxPVil-cre x IL-10 -/- mice help understanding carcinogenesis in the proximal colon, a cancer hotspot in patients with Lynch syndrome or inflammatory bowel disease.
