Genome-wide RNA-Seq analysis of circular RNA under transient MCAO rat model conditions

The model of transient cerebral focal ischemia was used. It was realized on two-month-old male white rats of Wistar line. Animals were divided into two groups: "sham operation" and "ischemia". Each group contained 3 animals. Focal ischemia in experimental rats from "ischemia" was initiated by endovascular occlusion of right middle cerebral artery using monofilament for 90 minutes followed by reperfusion. Rats were withdrawn from the experiment 24 hours after the occlusion. Ischemic focus was localized in the cerebral subcortex. Genome-wide transcriptome analysis of circular RNAs level in rat subcortex from "sham operation" and "ischemia" was carried out using High-throughput RNA-sequencing (RNA-Seq). The RNAse R-treated fraction from total RNA was used for RNA-seq. Sequencing was carried out using an Illumina HiSeq 2500 instrument. At least 20 million reads (1/200 nt) were generated.
