Replication Data for: Discovery and characterization of a specific inhibitor of serine-threonine kinase cyclin dependent kinase-like 5 (CDKL5) demonstrates role in hippocampal CA1 physiology

Pathological loss-of-function mutations in cyclin-dependent kinase-like 5 (CDKL5) cause CDKL5 deficiency disorder (CDD), a rare and severe neurodevelopmental disorder associated with severe and medically refractory early-life epilepsy, motor, cognitive, visual and autonomic disturbances in the absence of any structural brain pathology. Analysis of genetic variants in CDD have indicated that CDKL5 kinase function is central to disease pathology. CDKL5 encodes a serine-threonine kinase with significant homology to GSK3, whichhas also been linked to synaptic function. Further, Cdkl5 knock-out rodents have increased GSK3 activity and often increased long-term potentiation (LTP). The data generated here is relevant to the development and characterization of a specific CDKL5 inhibitor that excludes cross-talk with other kinases, especially GSK3. We provide data from western blots and electrophysiological data related to LTP that characterize different CDKL5 inhibitors. Western blot images, obtained with standard imaging techniques, can be re-quantified by users. Electrophysiological data was obtained with specialized software (WINLTP, free source, or pCLAMP (Molecular Devices), obtained via licence) for obtaining recordings from neurons in brain slices using field recording or whole-cell patch-clamp recording techniques. These data sets can be re-analyzed by users. All details for obtaining this data and subsequent analyses, including statistics used, are in the published manuscript. For western blot data, statistical analysis and visualization is done via Sigmaplot. For electrophysiological data, raw traces or analyses from raw traces were uploaded into Sigmaplot 12.5 (obtained via license) for further analyses (including statistics) and visualization. Sigmaplot 12.5 (or later versions) will be required to view these files. Data are ordered with reference to manuscript figures. Within Sigmaplot, data is structured to indicate the date specific data were acquired, how they were analyzed and the statistical approaches used. There are no legal or ethical considerations for re-use of this data. Re-use of this data is limited to re-analyses; due to experimental conditions that may vary, this data should not be combined into other novel datasets.
