mRNA profiling on CART19 cells treated with GM-CSF receptor alpha blockade or GM-CSF blockade

RNA sequencing was performed on three biological replicates of CART19 alone, GM-CSFRA Ab with CART19, or GM-CSF Ab with CART19 T cells from normal donors were transduced with a replication-incompetent lentiviral vector expressing a second-generation chimeric antigen receptor (CAR) consisting of an anti-CD19 single chain variable fragment (FMC63) fused to 4-1BB and CD3ζ intracellular domains as previously described. CART19 were cultured alone or with GM-CSFRA Ab or GM-CSF Ab (10ug/mL). Conditions each stimulated with irradiated NALM6 (CD19 antigen) for 24 hours. RNA was then isolated for sequencing.
