Rooted plants material of E. nitens x E. globulus of 6 months old were used to four treatments. Treatment non-acclimated (NA) was applied 20/12 °C day/night temperatures and photoperiod was long day length (14 h). The second treatment cold acclimated before night frost, (CABF) was applied 8/4 °C day/night temperatures and photoperiod was short day length (10 h). In the third treatment cold acclimated after night frost (CAAF) was applied successive night frosts of -2 °C and photoperiod was short day length, were performed for 4 days. In the last treatment de-acclimation (DA) it was increased photoperiod to long day length (14 h) and 12/4 ° C day/night. A night frost of -6 °C during DA was applied to determine the plants survival and the percentage of leaf damage for assess their tolerance. From leaf samples, RNA material was isolated and sequenced for RNA-seq experiments. Then, the transcriptome obtained using the de novo assembly strategy with the Trinity software, was used to evaluate the cuticle genes involved in cold acclimatization, following differential gene expression approaches.
