NRDE2 interacts with transcription-associated and splicing factors and modulates gene expression and splicing

To investigate the function of Nuclear RNAi-Defective 2 (NRDE2) in gene expression, we performed RNA-seq following its transient knock-down. Differential expression analysis showed that loss of NRDE2 modulated the expression of thousands of genes. Overall design: HeLa cells tranduced with shRNAs targeting NRDE2 (shNRDE2) or a non-targeting control (shControl) were harvested 5 days post-transduction. Total RNA was extracted using TRIzol. Paired end sequencing of polyA+ selected mRNA (20M clean reads per sample, 125 bp reads) was carried out.
