Chuanmai 42 is a core cultivar widely used in Winter Wheat Breeding Program in China. It is characterized by desirable yield related traits including high fertile spikelet number per spike (FSN) due to its long spike. Zhongkemai 13F10 is a stable breeding line selected by our lab characterized by higher grain number per fertile spikelet (GNFS). A recombinant inbred line (RIL) population derived from a cross of ZKM13F10/CM42 was used for BSE-Seq analysis. Based on the FSN values obtained in six environments, lines were re-arranged, and the two tails (each 30 lines) were selected to bulk extreme pools. Two pools (FSN-H and FSN-L) were bulked using equal quantities (1 μg) of DNA from 30 individuals in the tails. Exon capture and sequencing of these four libraries (ZKM13F10, CM42, FSN-H, and FSN-L) were performed by Bioacme Biotechnology Co., Ltd (Wuhan, China, http://www.whbioacme.com).
