Intra-individual stability of the urine miRNA transcriptome was examined by investigating longitudinal changes over time in a cohort of patients with localized prostate cancer. Using training and validation cohorts, urinary miRNA biomarkers are characterized and validated their utility to identify aggressive prostate cancer. miRNAs are isolated from urinary cell sediments. Discovery cohort is composed of 22 samples from 10 patients and used to measure intra-individual stability. Training cohort that are composed of urine samples from 49 low-risk prostate cancer patients and 50 high-risk prostate cancer patients are used to identify urine biomarker to predict cancer aggressivity and generate a predictive model. The predictive model is validated using an independent validation cohort with 29 low-risk and 15 high-risk (based on NCCN-risk, 11 high-risk based on GS-risk) prostate cancer patients. Control samples are used for data normalization.
