Cucumber (Cucumis sativus, cv. Senyal, HiShtil, Israel) plants were cultivated under controlled conditions in a greenhouse covered with insect-proof netting (50 mesh). The plants were grown in soilless media bags containing compressed coco coir, 16 X 40 X 120 cm H:W:L (Growbags, Pelemix, Isreal). Plants were spaced 30 cm apart. A drip irrigation system was utilized to supply water containing fertilizer according to local commercial standards. To prevent infection from insect-borne plant viruses and insect infestations, the plants were sprayed once a week with a tank-mixture of two systemic insecticides: a 0.01% solution of CONFIDOR (imidacloprid-based, BAYER, Germany) and a 0.02% solution of VERIMARK (cyantraniliprole-based, DUPONT, Delaware, USA).CGMMV viral inoculum was obtained by grinding cucumber leaves from CGMMV infected plants maintained in the greenhouses at the Volcani Institute, Rishon LiZion, Israel. The leaves were grinded in sodium phosphate buffer (10 mM, pH = 7.0) as previously described by (Molad et al., 2021). Mechanical inoculation was performed by hand-rubbing sap on the three young leaves in each infected plant. This treatment was designed to recreate wounding and viral transmission during field work and harvesting. Infection efficiency was validated by monitoring leaf symptoms and with a PCR-based assay.
