EZH2-mediated epigenetic suppression of IFNa/b-STAT2 signaling enhances anti-tumor immunity in breast cancer [ChIP-Seq]

Knockdown of catalytic inhibiton of EZH2 stimulates STAT2 recruitment in ISG promoter regions in luminal breast cancer cells MCF7 cells treated with EZH2 inhibitor or transduced shEZH2 were cross-linked unsing formaldehyde, and sonciation was performed to fragment chromtain between 100-300 bp in size ragne using sonciatior. Chromatin was immunoprecipitated with antibodies conjugated protein G dynabeads, followed by washing and elution. ChIP DNA was reverse crosslinked overnight and purifed by phenol precipitation. 2ng ChIP DNA was used for library construction and sequenced with Illumina HiSeq 3000.
