Data from: Evaluating the Use of the Extracellular Flux Analyzer Assay to Determine Mitochondrial Effects of Pesticides with Zebrafish (Danio rerio)

This study used the zebrafish model to systematically test organismal and subcellular toxicity of 24 widely used pesticides by quantifying developmental deformities and mitochondrial dysfunction following exposure. We first performed a 96-hour developmental assessment to evaluate deformities following exposure to each individual pesticide. For each pesticide, testing began with an initial range of concentrations: 0 (control), 10, 100, 1000, 10000, and 25000 ppb. Survival was recorded daily. At 96 hpf, larvae were assessed for developmental deformities (presence/absence based on deformity type). These data can be found in the Survival_96hpf.csv and Development_96hpf.csv files herein. Refinements of test concentrations were made by repeating these methods until a concentration was identified for each pesticide that resulted in ≥80% survival and ≈20% developmental deformities. This concentration then became the highest concentration used for mitochondrial function assays. Mitochondrial function was assessed using the Agilent Extracellular Flux Analyzer, measuring oxygen consumption rates (OCR) with and without certain drugs that modulate different parts of the mitochondrial to determine various parameters. These data can be found here in both raw OCR values as well as files that have been processes to exclude outliers and make calculations of these parameters.
