Depression is a serious and common mental health condition that affects more than 280 million people worldwide. People with major depressive disorder (MDD) often experience long periods of low mood, loss of interest, sleep issues, and difficulty functioning in daily life. While there are many approved medications to treat depression, not everyone responds the same way. In fact, only about one in three people fully recover after trying the first antidepressant, and doctors currently choose treatments based on trial and error rather than knowing which medication will work best for each person.
We will carry out a large research study to better understand who benefits most from antidepressant treatments. To do this, we will combine individual-level data from many clinical trials that tested antidepressants in adults diagnosed with MDD. These trials include people aged 18 to 80 and meet strict standards for quality and reporting. We will only use trials that have shared anonymized participant data under approved data-use agreements.
By analyzing this combined dataset, we will look for patterns across thousands of people to identify which personal and clinical features—such as age, sex, severity of depression, or presence of anxiety—help predict whether someone is likely to benefit from a specific medication. These features may include factors like how long someone has had depression, how many times it has come back, whether they also have anxiety, and their use of other medications.
We will use a type of statistical method called individual participant data (IDP) meta-analysis, which allows us to analyze data from different studies as if it came from one big study. This helps us get more accurate and reliable results. We will also account for differences between trials, such as the way data was collected or the type of participants included.
The goal of our research is to support more personalized treatment decisions for people with depression. Our findings will help doctors choose the best treatment for each patient more quickly and with greater confidence. This can reduce suffering, improve quality of life, and lead to faster recovery. We also hope that our results will guide the design of future clinical trials to make them more efficient and informative.
