We used RNA-Sequencing to quantify the changes in global mRNA expression in wild-type as compared to SWELL1 (LRRC8a) knock down of human umbilical vein endothelial cells (HUVECs). Overall design: We treated HUVECs with adenoviruses containing either Ad5-mCherry-U6-hLRRC8A-shRNA to knockdown SWELL1 or Ad5-U6-scramble-mCherry as a control. These cell lines were grown in culture and RNA was extracted for sequencing under basal culture conditions. RNA was sequenced on an Illumina Hi-Seq at the University of Iowa Core Facility.
