L. monocytogenes encodes a predicted ABC transporter, which we named EslABC, in its genome (lmo2767-9). An L. monocytogenes eslB mutant is unable to grow on BHI plates containing 100 µg/ml lysozyme or 0.5M sucrose and 0.025/0.05 µg/ml penicillin,. but readily forms suppressors. These suppressors contain secondary mutations, which can suppress the growth defect of the eslB mutant. To identify these mutations, the genome sequence of a selection of eslB suppressors was determined by whole genome sequencing using an Illumina MiSeq machine and a 150 paired end Illumina kit. We could identify secondary mutations in genes murZ, reoM, prpC, walR, walK, ftsE, ftsX, cwlO or in the promoter region of the dlt operon.
