Transcriptome analysis of Arabidopsis roots across domains

We performed RNA-seq analysis using WT(Col-0) and ipt3/5/7 mutant across domains. The position of root tips in seedlings grown on solid 1Ã? MS medium was recorded at ZT16 from day 4 to day 8 after sawing to establish boundaries for domains III to VI. The boundary between domains I and II was set at the root tip position at ZT2 on day 8. For RNA and histological sampling, roots were quickly cut at the boundaries at ZT16 on day 9.
