通过对全国48家博物馆10余种类型的文物开展生物病害调研获得本课题研究对象——馆藏文物害虫及霉菌,通过构建包含图片库和基因序列库在内的馆藏文物生物病害数据库,搭建我国首个文物常见生物病害在线识别专家系统,快速实现菌种和虫种类别的在线识别。 通过生物显微镜对常见霉菌进行显微形态拍摄;通过超景深显微镜对害虫进行形态照片拍摄;通过对馆藏文物表面霉菌采样后,分离纯化提取DNA,经一代测序技术获得真菌ITS序列,并通过NBCI数据库比对后的菌种初步鉴定结果。 虫种库图片数据文件夹包含赤拟谷盗、皮蠹、嗜卷书虱、衣鱼、杂拟谷盗等5种常见文物害虫各部位特征图片223张,其中赤拟谷盗69张、皮蠹27张、嗜卷书虱48张、衣鱼42张、杂拟谷盗37张。 菌种库显微图片数据文件夹包含镰刀菌、毛壳菌属、木霉属、青霉属、曲霉属、枝孢属等6种常见文物霉菌显微形态特征图片数据6703张,其中镰刀菌属360张、毛壳菌属181张、木霉属745张、青霉属2765张、曲霉属1915张、枝孢属737张。 菌种库序列文件夹包含项目实施期间馆藏文物表面霉菌ITS序列数据467条菌种初步鉴定结果。

The research subjects of this study, namely pests and molds infesting collection cultural relics, were obtained through a biological disease survey covering more than 10 types of cultural relics from 48 museums across China. By constructing a biological disease database for collection cultural relics that includes an image library and a gene sequence library, we developed the first online recognition expert system for common biological diseases of cultural relics in China, which enables rapid online identification of fungal and insect species. Common molds were photographed for their microscopic morphological features using biological microscopes, while pests were photographed for their morphological characteristics using ultra-deep field microscopes. After sampling molds from the surfaces of collection cultural relics, DNA was isolated and purified, fungal ITS sequences were obtained via first-generation sequencing technology, and preliminary species identification was conducted through alignment against the NCBI database. The insect species image data folder contains 223 feature images of different body parts of 5 common cultural relic pests, including Tribolium castaneum, Dermestes beetles, Liposcelis bostrychophila, Lepisma saccharina, and Tribolium confusum, with 69 images for Tribolium castaneum, 27 for Dermestes beetles, 48 for Liposcelis bostrychophila, 42 for Lepisma saccharina, and 37 for Tribolium confusum respectively. The fungal species microscopic image data folder contains 6,703 microscopic morphological feature images of 6 common cultural relic molds, namely Fusarium, Chaetomium, Trichoderma, Penicillium, Aspergillus, and Cladosporium, with 360 images for Fusarium, 181 for Chaetomium, 745 for Trichoderma, 2,765 for Penicillium, 1,915 for Aspergillus, and 737 for Cladosporium respectively. The fungal species sequence folder contains 467 sets of preliminary species identification results derived from fungal ITS sequence data of molds collected from the surfaces of collection cultural relics during the project implementation period.
