This study explores the crucial role of ventrolateral pallial excitatory neurons (ExNs) in integrating cognitive and sensory information through reciprocal connections with the prefrontal cortex (PFC). The research identifies transcription factors SOX4, SOX11, and TFAP2D as key regulators of these neurons' development, identity, and PFC connectivity. In mouse models, the absence of SOX4 and SOX11 significantly reduces the size and connectivity of specific brain regions, highlighting the importance of these factors. The study also underscores the role of TFAP2D dosage in shaping BLC-PFC connectivity and related behaviors, offering insights into neuropsychiatric disorders.
