We used Hi-C technology to reveal the landscape of three-dimensional chromatin architecture for the brains of AD patients. By comparing to that of age-matched and younger individuals without dementia, we reported that AD patients were more affected during ageing and AD genesis. Differential interaction analysis indicated four class of long-distance interaction shift during AD genesis and ageing. Among them, feedback class was wide-spread. We reported a more detailed long-distance interaction pattern between active regulatory regions, especially within gene body of the same genes, and observed that most of known AD risk genes were under long-distance promoter-enhancer interactions.
