SPOP inhibitor stabilize STING protein and activate STING signaling in vitro, which may enhance anticancer immunity. We used single cell RNA sequencing (scRNA-seq) to analyze the effect of SPOP inhibitor on the composition of different types of infiltrated CD45+ immune cells in B16 melanoma tumors Overall design: B16 tumor-beraring C57BL/6J mice were daily treated with or without 60 mg/kg of SPOP-IN-6b from day 5 to day 13. On day 14, tumor-infiltrating CD45+ cells in B16 tumors were enriched through positive selection via anti-CD45 biotinylated antibody and streptavidin pulldown, followed by scRNAseq.
