The teosinte (Zea mays subspecies parviglumis) inbred line TIL11 is the ancestor of maize (Zea mays L.). In this study, we used single-molecule long-read sequencing of a teosinte inbred line, TIL11, to construct a draft genome and a reference transcriptome for teosinte. We sequenced the genome and transcriptome of TIL11 with single-molecule long-read sequencing and paired-end second-generation sequencing. The transcriptome data generated from five tissues. The leaf, seedlings underground, and seedlings aboveground were sampled at the V4 stage. The ear and tassel were sampled at the VT stage. The Illumina cDNA paired-end libraries were constructed with an insert size of 350 bp, and sequencing was performed on the Illumina HiSeq 4000 platform. The PacBio single-molecule real-time libraries were constructed with full-length cDNA and sequenced. DNA was isolated from leaf tissues at the V4 stage. Total genomic DNA was fractionated into 10-50 kb fragments and sequenced using the GridION X5 sequencer platform.
