The present study revealed that RANKL can synergize with IL-13 and IL-4 to promote intestinal tuft cell expansion. To further explore the mechanisms downstream of RANKL, we conducted RNA-seq analysis of intestinal organoids cultured from Vil-creERT2;Rank fl/fl mice with or without RANKL treatment. The present also determined that ILC3s-derived RANKL is required for optimal tuft cell-ILC2 circuit activation during helminth infection. To elicidate the adaptation and contribution of ILC3s in helminth infection, we conducted scRNA-seq analysis of intestinal lamina propria leukocytes isolated from mice with or without N. brasiliensis infection for 7 days.
