To provide a high throughput approach to monitor the prevalence of target-site resistance to herbicide in agricultural fields, we describe a PacBio amplicon workflow for pooled samples that can be easily adapted to any gene of interest, independently of the organism. In this proof-of-concept study, we sequenced with PacBio High Fidelity (HiFi) reads long-range amplicons (13.2 kb) encompassing the entire ACCase gene in pools of over hundred blackgrass (Alopecurus myosuroides) individuals for various Europe-wide (9) and in German (64) field populations.
