Acute myeloid leukemia (AML) is the most common form of leukemia in adults. Differentiation therapy targets the key pathologic features of AML, the blockade of differentiation in myeloid progenitor cells. This study screened 100 plant extracts native to South Korea to search for those with differentiation-inducing activity in AML. Of these, Corydalis incisa (Thunb.) Pers. (CIP) exhibited the highest efficacy.CIP induced myeloid differentiation, decreased cell viability, increased cell apoptosis, and cell cycle arrest in multiple AML cell lines. We performed Quantseq 3' mRNA seqencing to understand CIP-induced differentiation and identify potential targets of CIP using CIP-resistant cell line. Overall design: To identify genes involoved in Corydalis incisa (Thunb.) Pers. (CIP)-induced AML differentiation, we generated CIP-resistant HL-60 cells (HL-60-CIP).
