Neuropathic pain (NP) has a high incidence in the general population, is closely related to anxiety disorders, and has a negative impact on the quality of life. As a natural product, Cannabidiol (CBD) has been widely studied for its potential therapeutic effects on symptoms such as pain and depression (DP). However, the mechanism by which CBD improves neuropathic pain with depression is not fully understood. Therefore, we used multi-omics studies (transcriptomics combined with proteomics) to deeply explore the intersection genes of CBD treatment of NP and DP. Secondly, the core targets were screened by protein-protein interaction (PPI) network, gene ontology (GO), Kyoto Encyclopedia of genes and Genomes (KEGG) analysis, molecular docking and molecular dynamics simulation. Behavioral tests were performed to evaluate the effects of CBD intervention on pain and depression behavior in mice with spinal nerve ligation (SNL), and a dose-response curve was drawn. After determining the optimal intervention dose, the core targets and signaling pathways were verified by Western blotting (WB) and quantitative polymerase chain reaction (qPCR). Finally, we investigated the underlying mechanism of CBD by Nissl staining, immunofluorescence (IF) and transmission electron microscopy (TEM). This study aims to provide further theoretical and experimental basis for the discovery of CBD as a potential therapeutic agent. The experiments were divided into three groups: sham-operated group, SNL model group and CBD (5mg/kg, ip) intervention group.
