Title: Agroinfiltration of non-guided pathway for moth pheromones production with copper-inducible dCas9EV system in Nicotiana benthamiana leaves
Description: Two constructs were made that comprised the three genes required for moth pheromone production (AtrD11, HarFar and EaDAct/ATF1) under the regulation of the synthetic promoters created in T11. The constructs were first tested transiently in Nicotiana benthamiana leaves via agroinfiltration, together with a plasmid containing the dCas9EV system under the regulation of a copper-inducible promoter and the CUP2:Gal4 protein. Agroinfiltrated leaves were treated 3 days after infiltration with Cu2SO4 1mM or water for the controls. Leaf samples were collected 2 days after copper treatment and the pheromone production was assayed via GC-MS
