This project aimed to analyze the transcriptomic changes in sun and shade leaves of Acer japonicum (fullmoon maple) during autumn leaf senescence. This project included isoform sequencing (Iso-seq) analysis to obtain full-length cDNA sequences and RNA-seq analysis using DNBSEQ reads. Three leaves from the north-faced outer canopy (sun leaves), and another three leaves from the north-faced inner canopy (shade leaves) of an adult A. japonicum tree (height: approximately 10 m, age: 50 years old) grown in the arboretum of Hokkaido Research Center, Forestry and Forest Products Research Institute (43.0°N, 141.4°E; 180 m a.s.l.) were used for the total RNA extraction. The leaves were harvested on July 7, July 17, and September 7, 2021, for Iso-seq analysis and on September 7, September 28, October 5, and October 13, 2021, for transcriptome analysis.
