Pleurotus pulmonariusis a warm weather oyster mushroom species that presents tolerance to heat stress, however the mechanism of its response to heat stress is still not understood. In this study, we provide analyses of the transcriptome of P. pulmonarius when grown under heat stress (40ºC). Five mycelium RNA samples were collected at 0, 2, 4, 6 and 8h under 40ºC and sequenced. A total of 289,332,406 clean reads were assembled into 14,438 unigenes, of which 9,248 unigenes could be matched to public databases. A total of 2,474 unigenes were differentially expressed at least at onetime point at an early stage of heat stress. Further comparative analysis showed that 308 genes were up- or down-regulated at four time points of the stress treatment, including 105 induced genes and 203 repressedgenes, which were called general responsive differentially expressed genes (DEGs). These results provide the basis for mining genes associated with the early response to heat stress in P. pulmonarius and offer insights for exploring the molecular mechanisms of heat stress tolerance in mushrooms.
