Transcriptomic analysis for heat-treated Arabidopsis seedlings

This study investigates how protein homeostasis (proteostasis) is regulated during acquired thermotolerance in Arabidopsis thaliana. By integrating transcriptome profiling with protein-level analyses, we reveal the dynamic pathways that help plants maintain proteostasis under heat stress. Our findings provide new insights into how plants cope with abiotic stress and highlight the role of proteostasis mechanisms in stress resilience.To this end, we used five-day-old Arabidopsis seedlings and primed them by exposing them to 37degC for 90 min, followed by a recovery at 22degC for 90 min, and heat-triggering at 45degC for 45 min. The unprimed seedlings (naive) were directly exposed to 45degC for 45 min. Control seedlings were maintained at optimal growth conditions (22degC, 16h light and 8h dark) throughout the experiment. We collected the samples at desired timepoints and compared the transcriptome across conditions.
