To establish an in vitro system to accomplish normal follicle formation, gonads isolated from mouse fetuses at embryonic day 12.5 were cultured for 7 days and then the transcriptome analysis was performed. This revealed that estrogen signal was excessively activated in cultured ovaries compared with in vivo-derived ovaries. Addition of estrogen antagonist, 10 ?M ICI182780, to the medium resulted in recovering from abnormal follicle formation in vitro. Transcriptome analysis also showed that estrogen signaling pathway was not excessively activated by ICI182780 incorporation.
