Structural basis for binding of RILPL1 to TMEM55B reveals a lysosomal platform for adaptor assembly through a conserved TBM motif

The TMEM binding motif (TBM) in diverse cytosolic proteins enables binding to lysosome localized TMEM55B. Here we have designed peptide mimics of the TBM to measure the affinities and thermodynamics of the interactions with a cytosolic region,. approximately 80-166, of TMEM55B. The peptides, and in the case of OCRL, the PH domain, were injected into a cell containing the protein using an ITC200 calorimeter from Particular Sciences. Data were processed using Origin 7.0 software and the final prcessed files in OPJ format are provided here, for each titration.
