Comparative analysis of DNA extraction protocols for ancient soft tissue museum samples

We compare the performance of two DNA extraction protocols (a commercial and a custom laboratory aDNA method) on hair and skin samples from decades-old museum specimens to Iron Age archaeological material. While both methods could recover DNA from ancient soft tissue, the Lab method performed better overall in terms of DNA yield and quality, which we could trace primarily to the poor performance of the commercial binding buffer in recovering aDNA.
