Virus growth kinetics was determined in vitro using four cell lines (Vero E6, Vero, SW13 and BHK-21) at different multiplicity of infection values. After, we established a mutation accumulation experiment by performing ten rounds of serial passaging and cross-passaging, followed by whole genome sequencing and bioinformatic analysis to map growth-adaptive signature mutations.
