Evaluation of genetic inactivation of lysine deacetylases KDAC6 (HDAC6) and KDAC8 (HDAC8) versus treatment with KDAC-selective inhibitors

To investigate the specificity and biological impacts of KDAC-selective inhibitors, we treated wild-type HT1080 cells and cell lines containing point mutations that result in endogenous expression of a catalytically-inactive variant with KDAC6 and KDAC8 selective inhibitors Tubastatin A and PCI-34051. Expression profiling analysis was then performed on the treated cells vs. untreated wild HT1080 cells. Overall design: Comparative gene expression profiling analysis of RNA-seq data for inhibitor-treated HT1080 cells and variants containing inactivated KDACs, with 3 replicates.
