Currently, an effective mouse model for gastric adenocarcinoma, the most common histological subtype, is unavailable. Through Cre-LoxP system, we successfully established a primary orthotopic gastric adenocarcinoma within 2.5 months, with the intestinal type emerging as the most prevalent. A mouse-derived subcutaneous allograft (MDA) model was developed, and a cell line was established from MDA, with prominent malignant characteristics. The subcutaneous allograft was sensitive to chemo-immunotherapy. This genetically engineered mouse model of gastric adenocarcinoma is an invaluable tool for investigating the tumor microenvironment, exploring anti-tumor immune responses, and evaluating drug sensitivity in vivo.
