Microglia are considered as a heterogenous population in the brain tissues. We used single-cell RNA sequencing (scRNA-seq) to analyze the diversity of microglia in the rats with chronic cerebral hypoperfusion. Overall design: Young male Sprague-Dawley rats (300-350g, 8 weeks old) were obtained, and bilateral common carotid artery occlusion (BCCAO) method was adopted to establish CCH model and mimic vascular dementia. We split brain tissues of sham and CCH rats into gray matters and white matters after 4 and 8 weeks of modeling (abbreviated to 4W/8W SG, SW, CG and CW). Microglia were sorted by magnetic activated cell sorting (MACS) using CD11b/c microbeads and analyzed using scRNAseq. Gray or white matter single-cell suspensions sorted by MACS in each group were derived from 3 different rats and mixed into 1 sample.
