Effect of depletion of LRIG1 on polyclonal CD8+ tumor infiltrating T cells and Effect of depletion of LRIG1 on OT1 CD8+ tumor-infiltrating T cells

To investigate the function of LRIG11 in regulating the differentiation of polyclonal CD8 tumor infiltrating T cells during the response to B16melanoma. And, To investigate the function of LRIG1 in regulating the activaiton of ovalbumin specific OT1 CD8+ T cells during the response to B16 tumor stably expresss ovalbumin as surrogate tumor antigen. Overall design: 3 independent biological replicates of WT and 2 independent replicates of LRIG1 KO CD8 T cells were sorted from B16 melanoma tumor tissue, on day 19 post vaccine treatment: each sample is purifed from pooled 4-5 tumor tissues. And for OT1 set WT and LRIG1 KO OT1 T cells were co-transferred at 1:1 ratio into mice which were then inoculated with B16ova tumor. We then sorted OT1 T cells out of tumor tissues on day 7 post exposure to tumors. 3 independent replicates of WT OT1 and 3 independent replicates of LRIG1 KO OT1 T cells were isolated from B16ova tumor tissues: each sample is purified from pooled 2-3 tumor tissues
