Alloreactive CD4+ T cells play a central role in allograft rejection, We used single cell RNA sequencing (scRNA-seq) to analyze the cell states of alloreactive CD4+ T cells in periphery and allografts to better understand how alloreactive CD4+ T cells drive allograft rejection. Overall design: C57BL/6 (B6) mice received adoptive transfer of congenically marked naïve WT or Ldha-/- TEa (TCR transgenic) CD4+ T cells, specifically recognizing the BALB/c I-Eñ major alloantigen. Subsequently, these mice underwent transplantation of BALB/c hearts. After 7 days post-transplantation, we isolated WT TEa cells from both spleens and heart allografts, along with Ldha-/- TEa cells from the spleens, for single-cell RNA sequencing (scRNA-seq) analysis.
